New Methodological Approaches in The Development of Russian Live Attenuated Vaccine for Pandemic Influenza

چکیده
Avian influenza viruses remain a major pandemic threat. In response to this threat, a number of pandemic vaccines have been developed. The objective of this paper is to review and summarize data from preclinical and clinical evaluation of Russian live attenuated influenza vaccines (LAIVs) against pandemic influenza based on cold-adapted A/Leningrad/134/17/57 (H2N2) master donor virus (MDV). The described LAIVs consist of reassortant viruses of 6:2 and 7:1 genomic composition (6 MDV genes: 2 WT genes and 7 MDV genes: 1 WT gene, respectively). Despite the differences in their genomic composition (6:2 or 7:1), LAIV candidates of H5, H7 and H2 subtypes acquired temperature sensitivity, cold-adaptation, and attenuation for different animal models. In addition, they were safe and immunogenic for healthy adult volunteers. The collected data indicate that 7:1 reassortants carrying HA genes of potentially pandemic viruses and the remaining genes from the MDV might be preferable pandemic LAIV candidates.

Potentially Pandemic Live Influenza Vaccines Based on Russian Master Donor Virus are Genetically Stable after Replication in Humans
Stability of attenuating mutations is an important premise for live attenuated influenza vaccine (LAIV). The presence of multiple mutations in internal gene segments of live attenuated viruses contributes to the stability of their genomes and consistent phenotypic properties. This study describes the evaluation of results of clinical isolates obtained in Phase I clinical trials of three Russian...

Seasonal influenza continues to have a huge annual impact in the United States, accounting for tens of millions of illnesses, hundreds of thousands of excess hospitalizations, and tens of thousands of excess deaths. Vaccination remains the mainstay for the prevention of influenza. In the United States, 2 types of influenza vaccine are currently licensed: trivalent inactivated influenza vaccine ...
متن کاملChallenges and Perspectives toward Development of more Effective Influenza Vaccine
Influenza viruses continue to be a major health threat in human and bird populations. The improvements in formulation and production level of the current influenza vaccines are not sufficient to afford complete protection. The continuous antigenic drifts and emergence of endemic and zoonotic strains make influenza vaccine planning difficult. Concern about the emergence of new influenza pandemic...

During the past decade, a number of H5 subtype influenza vaccines have been developed and tested in clinical trials, but most of them induced poor serum antibody responses prompting the evaluation of novel vaccination approaches. One of the most promising ones is a "prime-boost" strategy, which could result in the induction of prompt and robust immune responses to a booster influenza vaccine fo...
